Gene copy-number changes and chromosomal instability induced by aneuploidy confer resistance to chemotherapy

چکیده

Summary Mitotic errors lead to aneuploidy, a condition of karyotype imbalance, frequently found in cancer cells. Alterations chromosome copy number induce wide variety cellular stresses, including genome instability. Here, we show that cells might exploit aneuploidy-induced instability and the resulting gene copy-number changes survive under conditions selective pressure, such as chemotherapy. Resistance chemotherapeutic drugs was dictated by acquisition recurrent karyotypes, indicating dosage play role driving chemoresistance. Thus, our study establishes causal link between aneuploidy-driven chemoresistance explain why some chemotherapies fail succeed.

DNA copy number aberrations associated with aneuploidy and chromosomal instability in breast cancers.

Biological characteristics of a tumor are primarily affected by its genomic alterations. It is thus important to ascertain whether there are genomic changes linked with DNA ploidy and/or chromosomal instability (CIN). In the present study, using fresh-frozen samples of 46 invasive breast cancers, laser scanning cytometry, array-based comparative genomic hybridization, and chromosome fluorescenc...

Chromosomal Instability, Aneuploidy, and Cancer

The link between aneuploidy and cancer has been recognized over a century ago (1). Abnormalities in chromosome copy numbers arise from persistent errors in chromosome segregation during cell division, a process known as chromosomal instability (CIN) (2). CIN is a principal contributor to genetic heterogeneity in cancer (3) and is an important determinant of clinical prognosis and therapeutic re...

Analysis of copy number changes suggests chromosomal instability in a minority of large colorectal adenomas.

We have examined chromosomal-scale mutations in 34 large colorectal adenomas (CRAs). A small number of changes (median = 2, IQR = 0-4) were found by array-comparative genomic hybridization (aCGH) in most tumours. The most common changes were deletions of chromosomes 1p, 9q, 17, 19, and 22, and gains of chromosomes 13 and 21. SNP-LOH analysis and pseudo-digital SNP-PCR analysis detected occasion...

Chromosomal instability and aneuploidy in cancer: from yeast to man.

Aneuploidy is frequently associated with disease and developmental abnormalities. It is also a key characteristic of cancer. Several model systems have been developed to study the role of chromosomal instability and aneuploidy in tumorigenesis. The results are surprisingly complex, with the conditions sometimes promoting and sometimes inhibiting tumour formation. Here, we review the effects of ...

Reply to Heng: Inborn aneuploidy and chromosomal instability.

It is with great interest that we read the letter by Heng (1), who kindly brought up the interesting topic of nonclonal-chromosome aberrations (NCCAs) in the context of our recent report on the relationship between inborn aneuploidy and chromosome instability (CIN). We fully agree with Heng that NCCAs are better proxies for CIN than clonal-chromosome aberrations (CCAs). In fact, we used the fre...
